Analyze your opponent. It is Misty, so be ready for a Staryu and Starmie.

Analyze your team. Charmander would not be a good choice, as Misty is a water Pokémon trainer. It is an obvious type of disadvantage. Did you pick up a Pikachu in the Viridian forest? If you did, this gym will be easy; water conducts electricity. For pickers of Bulbasaur, this one will be easy as well.

Train all your current party members to level 25 at least. The higher they are, the better.

Utilize the kick and punch! Outside of Mt. Moon you may have noticed two people. One teaches your Pokémon Mega Punch, the other Mega Kick. These are great moves to use against Misty.

If you lose, try training to level 30 or catch some new Pokémon, such as Zubat and Clefairy.

Pikachu does not evolve at a specific level. In order to evolve him, go to the tower at Celadon City. On around the 3rd floor, you will meet a guy who will sell you a thunder stone, which is needed to evolve Pikachu.

What Pokemon should I use if I started with Squirtle?

You should just use an Electric type Pokemon. If you've got a Pikachu, that would be great. If you weren't able to get a Pikachu, just level up your Squirtle to be really powerful, and then take Misty on with lots of Super Potions.
